Erick Morillo, Pete Tong, Judge Jules, Fergie, Rob da Bank. Chris Coco, and Above & Beyond will all DJ at the infamous club in Playa d'en Bossa.
The party is the centrepiece of a month-long series of planned Radio 1 broadcasts from the island.
Throughout August, Pete Tong's Essential Selection will be broadcast live from Café Mambo, and all Essential Mixes for the month will be broadcast every weekend from Ibiza's clubs.
Tong said: "It's a milestone for Radio 1 to return to Ibiza for its 10th year, and I'm delighted that I'll be coming live with my show from Café Mambo.
"The big weekend starting on Friday 12th August will be my highlight - it's going to be very memorable indeed," he added.
Judge Jules will be kicking off the 13th August party at 7pm with a live two-hour broadcast from Space, which will be followed by a documentary celebrating the station's 10 years on the White Isle.
Radio 1's executive producer of live events, Jason Carter, said over the ten years Radio 1 had held some of the best parties ever witnessed on the island.
"August will see us going back to our Ibiza roots by broadcasting from Café Mambo, then culminating in a big club night at Space," he said.
"We plan to give the clubbers and listeners more than ever, with some of the best DJs in the world alongside some classics from our best Ibiza parties over the years."
Radio 1 also said an off-air free beach party at Café Mambo on Saturday 13th August was being planned from 3-7pm, although a line-up hasn't been confirmed.
